Large scale plasmid production is the step between a plasmid that works on the bench and a plasmid that goes into cells at scale, into a viral vector run or into a clinical material. The chemistry is the same as a miniprep; the difference is grade, documentation, endotoxin control and volume. The three grades and who needs them

Research grade is a clean, sequence-verified plasmid at milligram scale, endotoxin reduced, for transfection and in vitro work. High quality or transfection grade adds tighter endotoxin limits, a supercoiled fraction specification and fuller testing, and is what viral vector production and large cell-culture campaigns need. GMP grade is made under a quality system with batch records, released against a specification, and is the only grade acceptable as a starting material for clinical products. The grades differ more in documentation and testing than in the DNA itself.

What the certificate of analysis has to show

Read the certificate before the price. It should report concentration and total yield, purity by absorbance ratio, the supercoiled fraction by gel or capillary electrophoresis, endotoxin, residual host protein and RNA, and confirmation of identity by restriction digest or sequencing. For higher grades, sterility and residual antibiotic appear too. A vendor who cannot show a sample certificate for the grade you are buying is quoting a grade in name only.

Scale, yield and lead time

Yield per litre of culture depends on the plasmid: copy number, size and any toxic element all move it. That is why vendors quote by delivered milligrams rather than by culture volume, and why an unusual plasmid gets a feasibility run before a firm price. Lead time rises with grade because the testing does, and GMP batches are scheduled into a manufacturing slot rather than a queue. Ask for the lead time at your grade in writing and ask what a repeat batch will cost, since the second batch is where the real relationship price appears.
