Test tube selection: what a test tube is made of, how it is capped, and why rack and rotor compatibility decides the order
Test tubes are ordered on price and regretted on fit. The dimension that matters is rarely the volume printed on the box but the outside diameter and the length, because those decide whether the tube fits your racks, your heating block, your rotor and your reader. The specification that actually matters
- Outside diameter and length before volume. Racks, blocks, rotor adapters and readers are built around external dimensions. Check the diameter and length against everything the tube must sit in before ordering, because two tubes of the same nominal volume from different suppliers frequently differ enough to matter.
- Glass or plastic. Borosilicate takes direct heat, resists solvents and can be flame sterilised; polypropylene survives centrifugation and freezing and does not shatter; polystyrene is clear and cheap and is limited to ambient aqueous work. Each choice rules something out, so pick from the protocol.
- Round, conical or flat bottom. Conical bottoms concentrate a small pellet and make it visible; round bottoms mix better and suit culture; flat bottoms stand unaided and suit reading. The shape is a protocol decision that also determines whether the tube needs a rack to stay upright.
- Closure type. Screw caps seal reliably and are slow; push caps are fast and can pop under pressure or in a freezer; septum caps allow sampling without opening. For anything frozen, shipped or heated, the closure is the part that fails, so specify it deliberately.
Sterility, certification and lot control
Sterile tubes are supplied irradiated or otherwise treated and certified in intact packaging. For molecular work, suppliers also certify freedom from nucleases and from amplification-inhibiting contamination, and those certifications are separate from sterility; if your assay depends on one, ask for it by name on the order.
Where results feed a regulated process, ask whether the supplier provides lot traceability and retains certificates, because reconstructing that after a deviation is far harder than requesting it up front.
Centrifugation and freezing
Tubes carry a maximum rated force, and it depends on the adapter supporting them as much as on the tube. A tube rated in a snug adapter will fail unsupported, which is the usual cause of a tube failure in a rotor.
For freezer storage, check the rated low temperature and whether the closure stays sealed through a freeze and thaw. Tubes that pass at a domestic freezer temperature do not necessarily pass in ultra-low storage or in vapour phase.
Common questions
- What is a test tube made of?
- Usually borosilicate glass, polypropylene or polystyrene. Borosilicate takes heat and solvents, polypropylene takes centrifugation and freezing, and polystyrene is a clear low-cost option for ambient aqueous work.
- Can test tubes be autoclaved?
- Borosilicate and most polypropylene can, within the manufacturer's stated limits. Polystyrene cannot. Closures often have a lower limit than the tube, so check both.
- What is the difference between a test tube and a centrifuge tube?
- A centrifuge tube is rated for a maximum relative centrifugal force in a specified adapter and is usually conical with a sealing closure. A general test tube carries no such rating and should not be spun hard.
- Why do my tubes not fit the rack?
- Nominal volume does not fix external dimensions. Racks, blocks and rotor adapters are built for a diameter and length, and suppliers differ. Check both dimensions against every holder the tube must enter.
- Do I need certified nuclease-free tubes?
- For RNA work and sensitive amplification, yes, and the certification is separate from sterility. For general chemistry and culture it is an unnecessary cost.
