Purification platforms differ mainly in pressure and in how fractions are decided. Flash systems run fast at low pressure with disposable cartridges and suit synthetic chemistry; preparative liquid chromatography runs at pressure with reusable columns and finer control. Extraction avoids the column entirely when the chemistry allows.

Choosing and sizing
- Size from the loading, not the volume. How much can be loaded before resolution degrades sets the throughput. Measure it on an analytical column and scale by stationary phase mass rather than by column dimensions alone.
- Decide disposable cartridge against packed column. Cartridges remove packing variability and cleaning, and cost more per run. Reusable columns are cheaper per run and require packing skill and a cleaning regime. Volume decides.
- Pack properly or buy packed. A poorly packed bed channels, and the separation never recovers. Frits, packing technique and bed compression determine whether a self packed column performs, and for critical work a packed column from a supplier is the safer economy.
- Set fraction collection on a real trigger. Threshold, slope or time, chosen against the actual chromatogram, with an analytical method ready to check the fractions. Collecting blind and analysing later wastes material.
- Consider extraction before chromatography. Where the target and the impurity differ in partition behaviour, liquid or solid phase extraction is far cheaper and faster than a column. Try it first for simple separations.
Fractions are the product
A purification run is judged on how much pure material was collected, which depends on where the fractions were cut and how quickly they were analysed. A beautiful chromatogram with pooled impure fractions is a failed run.
Have the analytical method ready before the preparative run, and decide the pooling rule in advance.
Solvent dominates the cost
At preparative scale, solvent purchase and disposal frequently exceed everything else, and methods that use less are worth developing for that reason alone.
Include solvent volume in the method comparison. It is routinely omitted and it decides the economics.

- Flash for fast, lower resolution separations of synthetic mixtures at low pressure. Preparative liquid chromatography where resolution, reproducibility and detection control matter and the material justifies the cost.
- Are disposable cartridges worth it?
- For varied, occasional separations, yes, because they remove packing and cleaning variability entirely. At sustained volume the consumable cost favours a reusable packed column.
- Why does my self packed column channel?
- Uneven packing, bed compression or a damaged frit. All three produce a separation that cannot be recovered by method changes, and repacking is the remedy.
