Choosing and qualifying the kit
- Match the standard range to what you expect. Most cytokines circulate at very low picogram levels in health and rise sharply in disease, so a kit whose range brackets healthy values may saturate in patients and vice versa. Pick the range that puts your expected values in the middle of the curve where precision is best.
- Establish what the kit detects. Free cytokine, total including that bound to soluble receptors, or something in between depends on the antibody pair and on whether a dissociation step is included. Two kits measuring the same cytokine can legitimately give values that differ several-fold for this reason.
- Fix the pre-analytical protocol and follow it exactly. Tube type, time to centrifugation, temperature and freeze-thaw all move cytokine results, and several are released from leucocytes and platelets during clotting. Serum and plasma are not interchangeable and the anticoagulant matters.
- Qualify the kit in your own matrix. Spike a known amount into your actual sample type and recover it, and check that a serial dilution back-calculates consistently. A kit validated in serum used on culture supernatant or lysate is unvalidated until you show otherwise.
- Decide single analyte or multiplex deliberately. A multiplex panel measures many cytokines from one small volume and brings cross-reactivity and a compromise buffer with it. For one analyte that matters, a dedicated kit is usually more accurate; for a broad survey a panel is the only practical route.
Running the plate so the numbers survive review
Randomise samples across the plate rather than loading groups in blocks, because edge effects and wash gradients otherwise become group differences. Put a bridging control on every plate in a multi-plate study.
Run standards and samples in duplicate and look at the duplicates rather than only the mean. Disagreeing duplicates indicate a pipetting or washing problem that the mean conceals.
Interpreting a cytokine result
Circulating cytokine concentrations are dynamic over hours and vary with time of day, recent activity and infection. A single measurement is a snapshot, and conclusions about a group need enough subjects to see past that variability.
Report the kit, the lot, the matrix and the handling protocol in the methods. Most disagreements between published cytokine values come down to one of those four rather than to biology.
Kit against a custom assay
A kit arrives qualified and costs more per sample. Building an assay from a matched antibody pair is cheaper at volume, takes real development time and gives control of supply, which matters for a study running over years.
Whichever route, bank a single lot of the critical reagents and the standard for the study. A standard from a different lot shifts the whole curve, and that shift is indistinguishable from an effect.
