A custom vector order is a small manufacturing project. The packaging system decides the safety profile and what can be carried, the titre method decides whether two batches can be compared, and the purity decides whether primary cells survive the preparation.

Specifying a custom batch
- State the packaging system and generation. Later generation systems split the packaging functions across more plasmids and reduce the risk of a replication competent event. The generation is a safety statement and belongs in the order and in the biosafety application.
- Choose transient or integrating deliberately. An integrating vector gives lasting expression and inserts into the genome. A non integrating adenoviral vector gives high, transient expression without insertion. The experiment's duration decides.
- Name the titre method and the cell line. Functional titre on a cell resembling yours is what predicts transduction. Physical particle counts are much higher and do not. Fix it in the order.
- Specify purity for the target cells. Crude supernatant is adequate for robust lines and toxic to primary cells. Concentration alone concentrates the medium components too, so purification rather than concentration is what primary work needs.
- Order the control in the same production. An empty or non targeting vector from the same run shares the preparation and its contaminants, which is what a control has to do.
- Have the approvals before the delivery. Institutional biosafety approval, containment level and waste route, agreed and documented. This is administrative and it is the commonest cause of a delayed start.
Generation is a safety statement
Splitting packaging functions across separate plasmids reduces the chance of recombination producing a replication competent particle. Which system was used is therefore part of the risk assessment and not a technical detail.
Record it in the biosafety application and on the vial. A vector of unknown provenance is difficult to assess and difficult to justify.
Comparability between batches
Two batches of the same construct differ in titre, in purity and sometimes in behaviour. Where an experiment spans batches, that variation has to be handled by design rather than assumed away.
Order enough for the whole experiment where possible, and where not, bridge the batches with a common sample.

- Concentrate or purify?
- Concentration raises titre and also concentrates medium components, serum and cellular debris. For primary and sensitive cells, purification is what removes the toxicity, and concentration alone frequently makes it worse.
- Integrating or transient vector?
- Integrating for lasting expression through division; adenoviral or other non integrating routes for high, short lived expression without insertion. Duration of the experiment usually decides.
- What has to be in place before delivery?
- Institutional biosafety approval covering the vector and the insert, the containment level, a waste route and trained handlers. All of it is administrative and all of it takes longer than production.
