A compound microscope stacks two magnifying stages, an objective and an eyepiece, to reach magnifications a single lens cannot, and it is the instrument almost every laboratory means by microscope. Two quotations at similar prices can deliver very different instruments, and the difference is almost never in the headline magnification. This page covers what to check and in what order.

The checks that matter

  1. Objective correction and numerical aperture. Numerical aperture rather than magnification sets what the instrument can resolve, and the correction class sets how flat and colour-true the field is. A quotation that lists magnifications without numerical apertures and correction classes is withholding the specification that decides image quality.
  2. The condenser is half the optical path. An adjustable condenser with an iris, properly set up, is what produces contrast and resolution in transmitted light. Instruments sold with a fixed condenser are cheaper and cannot be aligned properly, and no objective compensates for that.
  3. Illumination quality and stability. Even, adjustable illumination with a stable colour temperature matters for imaging and for hours of comfortable viewing. Light emitting diode sources are now standard and vary considerably in evenness and in whether intensity changes shift colour.
  4. Head, eyepieces and field number. A binocular or trinocular head with adjustable interpupillary distance and dioptre correction is worth specifying, and the eyepiece field number determines how much of the image you actually see. A large field number with mediocre objectives shows more of a poorer image.
  5. Mechanical quality. Stage movement, focus feel and absence of drift are what make an instrument pleasant over years, and they are the things a brief demonstration reveals immediately. Ask to operate it rather than to watch it operated.

Compound against stereo

A compound instrument gives high magnification of thin, transmitted-light specimens and a very shallow depth of field. A stereo or dissecting instrument gives low magnification, a large working distance and a three-dimensional view of solid objects. They answer different questions and a laboratory doing both needs both.

Buying a compound instrument for dissection work is the most common mismatch, and it is obvious within a day of delivery.

Imaging, if you will do it

A trinocular head and a camera port are far easier to specify at purchase than to add later, and the camera should be matched to the objective's resolution rather than chosen for pixel count. An oversized sensor behind a modest objective records empty magnification.

Decide whether images are for documentation or measurement. Measurement needs a calibration slide and a recorded scale for each objective, which is a small discipline that makes the images usable later.
