Most ELISA disappointments are range and matrix problems rather than kit quality. A target present at grams per litre and a target present at picograms per millilitre need completely different kits, and a kit validated in serum may behave differently in culture medium or in lysate. Choosing and qualifying the kit

  1. Start from the expected concentration, not the target name. Albumin in serum is one of the most abundant proteins there is and needs enormous dilution; a neurotrophin or a cytokine sits at the other end of the scale. Pick the kit whose standard curve brackets your expected values in the middle of its range, where precision is best.
  2. Check the matrix the kit was validated in. Serum, plasma with a named anticoagulant, culture supernatant, lysate and urine are different matrices and each can interfere differently. A kit validated in serum used on culture medium is an unvalidated assay until you show otherwise.
  3. Run spike recovery and linearity of dilution on your own samples. Spike a known amount into your matrix and recover it, and dilute a high sample serially and check the back-calculated values agree. Recovery within roughly eighty to a hundred and twenty percent and parallel dilution are the two tests that qualify a kit for your sample type.
  4. Decide what the kit actually detects. Free against total, mature against precursor, and whether a binding protein in the sample masks the target. For several cytokines and neurotrophins this single question explains most of the disagreement between published values.
  5. Fix the handling before the first plate. Tube type, time to centrifuge, storage temperature and the number of freeze-thaw cycles all move results, and several cytokines are released from cells during clotting. Write the pre-analytical protocol down and follow it for every sample in the study.

Reading the standard curve properly

Fit the curve with the model the kit specifies, usually a four parameter logistic, and do not force a straight line through the middle. Values read from the flat ends of the curve carry far more error than the coefficient of variation suggests.

Run the standards in duplicate and look at the duplicates rather than only the mean. A plate where duplicates disagree has a pipetting or a washing problem, and the mean hides it.

Plate effects that look like biology

Edge wells evaporate and incubate differently, and an uneven wash leaves background in a pattern. Randomise sample position across the plate rather than loading groups in blocks, or a plate effect becomes a group difference.

Where a study spans several plates, put a bridging control on every plate. Without it, plate to plate variation cannot be separated from the effect you are measuring.

Kit against custom pair

A kit is faster, comes qualified and costs more per sample. Building an assay from a matched antibody pair is cheaper at volume and takes real development time, including choosing the pair, the blocking and the buffer.

For a long study at scale, the custom route usually wins on cost and on control of supply. For a handful of samples it almost never does.

Common questions

How do I choose between two human elisa kits for the same target?
Compare the standard range against your expected concentration, the validated matrices, the published spike recovery and dilution linearity, and what the antibody pair detects. Price rarely separates them usefully; range and matrix almost always do.
Why do two kits give different values for the same sample?
Different antibody pairs see different forms, and different standards are calibrated differently. Values from two kits are generally not interchangeable, which is why a study should use one kit and one lot throughout.
What does spike recovery tell me?
Whether your matrix interferes. Adding a known amount of standard to your sample and recovering roughly what you added shows the matrix is not suppressing or enhancing the signal; a recovery well outside that says it is.
Can I use a serum kit on cell culture supernatant?
Only after qualifying it. Medium contains serum components, phenol red and additives that can interfere, so run spike recovery and dilution linearity in your actual medium before trusting the numbers.
